Production and Detection of Virus - free Lily Plants by Shoot Tip Culture and Virazole Treatment of bulbils

Abstract
This study was conducted for the production of virus-free lily stocks by an application of shoot tip culture technique, virazole treatment and tissue blotting. The percentage of virus-free plants through shoot tip culture averaged 95% for all lily symptomless virus (LSV),cucumber mosaic virus (CMV) and tulip breaking virus (TBV). The number of detection times using tissue blotting to get virus-free plants was three times, once after two months of shoot tip culture, the second time after two months of subculture, and the third time after four months of the subculture. For using bulbils for LSV and CMV-free bulbs production of lilies, adequate concentration of virazole by spray on bulbils was 100 §·/L. In vitro culture, it was 20 §·/L. Both methods were highly effective for eradication of the viruses. In case of low concentration virazole treation in vitro, there were high numbers of small bulblets and their growth was good.
